Cooling of porcine semen in an extender supplemented with isoespintanol

ABSTRACT: Spermatozoa experience oxidative, osmotic, chemical, and thermal stresses when cooled, which degrade the quality fertilizing capacity of cells. Adding antioxidants to sperm extender mitigates these alterations. This study evaluated effect isoespintanol (ISO) on boar semen subjected cooling. Fifteen ejaculates from five boars (Susscrofadomestica) were extended in Beltsville thawing solution (BTS) supplemented with 0 µM (control), 5 (ISO5), 10 (ISO10), 15 (ISO15), 20 (ISO20), 25 (ISO25), 30 (ISO30) ISO, then cooled for days at 16 °C. Sperm kinetics, total motility (TM), progressive (PM) every 24 h using an IVOS computer-assisted analysis (CASA) system. On day 1 cooling, a hypoosmotic test, spectrofluorometry, flow cytometry performed evaluate following: membrane functionality, measured as function swelling (HOS); antioxidant (TAC); reactive oxygen species (ROS); mitochondrial potential (Δ¥M). Regression comparison means Duncan test performed. The ISO added had slight impact motility, evidenced by reduction TM cooling (but not prior) addition ISO.
